To exclude tags, there is an exclamation mark for each tag in the filter input above your link list.

Excluding a tag from search is all about getting a targeted hit list on search - improving the quality of the search results.
I may know I've got links on a topic somewhere, or I may not be sure so I want to find out what I've got.
Sometimes specifying a tag to search for generates a short list, at least a short enough list. You can look through that list.
But often I've got a huge list of hits on that tag, but that are still many tags that intersect with that tag and I'm not sure which ones might apply - but I know which ones don't.
So if I can search for tag - omitting other tags - I can remove 100s of hits from the search and get a reduced list to look through for what I'm looking for.
For example, I'm looking for someting on JavaScript so it'll be tagged JavaScript, but that's a lot of hits. Half my library is all about JavaScript. But I know it's not DOM, or frameworks, or libraries, which also covers a lot of JavaScript links. So looking for JavaScript minus DOM minus frameworks minus libraries will produce a vastly reduced list.
I'd probably want to then text search within that limited scope as well.
